Molecular Changes Induced by the Bisphenol A Sertoli Cell Biology Assay
Bisphenol-A BPA is well known as one of the endocrine disrupting and testicular toxic chemicals. In the present study, we determined whether BPA caused cell injury due to impaired mitochondria and overproduction of ROS. Cellular ROS production, mitochondrial ATP synthase activity and concentration were examined. We, To study the role of the NR5A nuclear receptor in the testis after sex determination, we analyzed mice lacking NR5A Sertoli SC cells from embryonic day E 13. Ablation of Nr5a, characteristic gene expression of SC identity, e.g. Sox Amh, caused SC death. The MTT assay for MBP in HSeC Sertoli cells showed that µM doses further inhibited cell viability h of exposure (Fig. 1-A ). When cells were exposed to selected doses of BPA, it was observed that µM doses further inhibited cell viability after both exposure periods (Fig. 1-B). Based on MTT, we reported that BPS exposure significantly promoted breast tumor proliferation and deterioration in a non-monotonic dose-response manner. The mechanisms were investigated by molecular biology and mass spectrometry-based lipidomics, proteomics and imaging. BPS exposure induced the spatially intratumoral heterogeneity of Pereira et al. 2004 demonstrated that Sertoli cells are affected by Cr V and that these cells have a large number of vesicles and vacuoles and that Cr V causes disruption of the blood. An important finding was that in primary human Sertoli cells, BPA exposure significantly decreased cell viability, expression levels of occludin, ZO-1, β-catenin and AR at a µM dose. Background. The testes are intended to serve two important roles: producing testosterone steroidogenesis and maintaining germ cell development. These functions are supported by testicular somatic cells, Sertoli SC cells, located in the seminiferous tubules of the testes 2, 3. Sertoli cells are considered most, particularly in the Sertoli cell, ruptured vesicles could be found in the multivesicular nuclear body. Conclusion: BPA treatment at low concentration tends to drive spermatogenic and Sertoli cells to apoptosis, whereas higher concentration tends to drive spermatogenic and Sertoli cells to necrosis. Therefore, this study showed. Sertoli cells are morphologically and functionally unique somatic cells that play a specialized role in supporting spermatogenesis. These cells are known to cyclically change their gene expression based on continued alteration of seminiferous epithelial stages, although the mechanism of regulation is largely unclear.
